The DNA of the diploid cells was measured in th G0/G1 (G1 phase). In this stage the cell sows down the growth and prepares itself for itself for mitosis. The chromosomes become condensed and are clearly visible during this state.

DNA content will be doubled in Anaphase. The cells will contain four sets of chromosomes during this stage.

At the beginning of G1 the diploid cell has two complete sets of chromosomes inherited from its parent cell. Each chromosome is unreplicated and thus consists of the DNA content of a diploid cell. In S phase the DNA is replicated and the content of DNA is doubled. Followed by metaphase and anaphase where the DNA content remains the same ( i.e. doubled).
